Most species of Amazonian snakes have wide geographic distributions. However, local environmental factors influence the formation of assemblages in different localities. In this study, we investigated the composition of the assemblage and the effect of environmental variables on the distribution of the species inhabiting an upland forest in the Experimental Farm area of the Federal University of Amazonas in Manaus, Brazil. Data collection was carried out in 24 standardized plots. Each plot was sampled four times between July 2015 and April 2017 by active search method. We recorded 83 individuals from 29 species belonging to six families. The richness in the study area corresponded to 78% of the snake species and 100% of the families previously recorded for Manaus. As observed in other localities, the most abundant species was the Amazonian lancehead (Bothrops atrox). Multiple linear regression models did not detect any effect of environmental variables on species richness and abundance of individuals. However, quadratic polynomial regression models revealed that intermediate canopy opening percentages positively influence the richness and abundance of snakes. It is possible that the result is related to a tradeoff between the thermoregulation behavior of these animals and to their susceptibility to predation.

DNA barcoding is a simple technique used to develop a large-scale system of classification that is broadly applicable across a wide variety of taxa. DNA-based analysis of snake venoms can provide a system of classification independent of currently accepted taxonomic relationships by generating DNA barcodes specific to each venom sample. DNA purification from dried snake venoms has previously required large amounts of starting material, has resulted in low yields and inconsistent amplification, and was possible with front-fanged snakes only. Here, we present a modified DNA extraction protocol applied to venoms of both front- and rear-fanged snakes that requires significantly less starting material (1 mg) and yields sufficient amounts of DNA for successful PCR amplification of regions commonly used for DNA barcoding. Atractaspis snake species are enigmatic in their natural history, and venom effects are correspondingly poorly described. Clinical reports are scarce but bites have been described as causing severe hypertension, profound local tissue damage leading to amputation, and deaths are on record. Clinical descriptions have largely concentrated upon tissue effects, and research efforts have focused upon the blood-pressure affecting sarafotoxins. However, coagulation disturbances suggestive of procoagulant functions have been reported in some clinical cases, yet this aspect has been uninvestigated. We used a suite of assays to investigate the coagulotoxic effects of venoms from six different Atractaspis specimens from central Africa. The procoagulant function of factor X activation was revealed, as was the pseudo-procoagulant function of direct cleavage of fibrinogen into weak clots. The relative neutralization efficacy of South African Antivenom Producer's antivenoms on Atractaspis venoms was boomslang>>>polyvalent>saw-scaled viper. While the boomslang antivenom was the most effective on Atractaspis venoms, the ability to neutralize the most potent Atractaspis species in this study was up to 4-6 times less effective than boomslang antivenom neutralizes boomslang venom. Therefore, while these results suggest cross-reactivity of boomslang antivenom with the unexpectedly potent coagulotoxic effects of Atractaspis venoms, a considerable amount of this rare antivenom may be needed. This report thus reveals potent venom actions upon blood coagulation that may lead to severe clinical effects with limited management strategies.

Snake bile is one of the most expensive traditional Chinese medicines (TCMs). However, due to the complicated constitutes of snake bile and the poor ultraviolet absorbance of some trace bile acids (BAs), effective analysis methods for snake bile acids were still unavailable, making it difficult to solve adulteration problems. In present study, ultrahigh-performance liquid chromatography with triple quadrupole linear ion trap mass spectrometry (UHPLC-QqQ-MS/MS) was applied to conduct a quantitative analysis on snake BAs. The mass spectrometer was monitored in the negative ion mode, and multiple-reaction monitoring (MRM) program was used to determine the contents of BAs in snake bile. In all, 61 snake bile from 17 commonly used species of three families (Elapidae, Colubridae and Viperidae), along with five batches of commercial snake bile from four companies, were collected and detected. Nine components, Tauro-3α,12α-dihydroxy-7-oxo-5ß-cholenoic acid (T1), Tauro-3α,7α,12α,23R-tetrahydroxy-5ß-cholenoic acid (T2), taurocholic acid (TCA), glycocholic acid (GCA), taurochenodeoxycholic acid (TCDCA), taurodeoxycholic acid (TDCA), cholic acid (CA), Tauro-3α,7α-dihydroxy-12-oxo-5ß-cholenoic acid (T3), and Tauro-3α,7α,9α,16α-tetrahydroxy-5ß-cholenoic acid (T4) were simultaneously and rapidly determined for the first time. In these BAs, T1 and T2, self-prepared with purity above 90%, were first reported with their quantitative determination, and the latter two (T3 and T4) were tentatively determined by quantitative analysis multi-components by single marker (QAMS) method for roughly estimating the components without reference. The developed method was validated with acceptable linearity (r2≥0.995), precision (RSD<6.5%) and recovery (RSD<7.5%). It turned out that the contents of BAs among different species were also significantly different; T1 was one of the principle bile acids in some common snake bile, and also was the characteristic one in Viperidae and Elapidae; T2 was the dominant components in Enhydris chinensis. This quantitative study of BAs in snake bile is a remarkable improvement for clarifying the bile acid compositions and evaluating the quality of snake bile.

Um inventário estruturado de serpentes foi realizado na Base Operacional Geólogo Pedro de Moura (BOGPM), localizada na Bacia Petrolífera de Urucu, Município de Coari, Amazonas, nos anos de 2003, 2004 e 2007. Nas quatro expedições realizadas (51 dias de coleta) foram registradas 47 espécies de serpentes, pertencentes a sete famílias e 33 gêneros. Foram utilizados quatro métodos complementares de amostragem de serpentes: armadilha de interceptação e queda, encontros ocasionais, procura limitada por tempo a pé e procura limitada por tempo de carro. Das 47 espécies coletadas, Liophis reginae (n= 14), Philodryas viridissima (n= 9), Philodryas boulengeri (n= 7) e Oxybelis fulgidus (n= 7) foram as mais abundantes em toda região. O maior número de espécies e espécimes foi registrado pela procura limitada por tempo de carro (52,8%). Estudos anteriores indicam que as localidades ao sul do Rio Amazonas (como região Leste do Pará, Usina Hidrelétrica de Tucuruí, Estado do Pará, e de Samuel, Estado de Rondônia) apresentam maior riqueza quando comparadas às regiões ao norte do Amazonas (como Município de Manaus, Reserva do INPA-WWF e Usina Hidrelétrica de Balbina, Estado do Amazonas). Desta forma, é possível inferir que o levantamento das serpentes da região de Urucu ainda não esteja completo, sendo necessário um maior esforço de coleta para que novos registros sejam adicionados para a área.


A structured snake inventory was carried out at the Base Operacional Geólogo Pedro de Moura (BOGPM), located at the Urucu Petrol Basin, Municipality of Coari, Amazonas, during 2003, 2004 and 2007. Throughout four sampling expeditions (51 collection days), seven species of snakes, belonging to seven families and 33 genera, were recorded. Four complementary sampling methods were used: pitfall traps with drift fence, occasional encounters, time-limited search on foot and time-limited search by car. Of the 47 collected species Liophis reginae (n= 14), Philodryas viridissima (n= 9), Philodryas boulengeri (n= 7) and Oxybelis fulgidus (n= 7) were the most abundant in the region. The highest number of species and specimens was obtained by the time-limited search by car (n=52.8%). Previous studies indicate that localities south of the Amazonas river (such as the eastern region of Pará and Tucuruí, state of Para, and Samuel Hydroelectric Plants, state of Rondônia) present a greater richness when compared to those north of the Amazonas river (such as the mucipality of Manaus, INPA-WWF Reserve and Balbina Hydroelectric Plant, state of Amazonas).Thus, it is possible to infer that the snake inventory in Urucu region is not yet complete, requiring more sampling efforts so that new records can be obtained for the area.
